My name is Elizabeth Gardner and I have been the Program Director for the Graduate Studies in Forensic Science at the University of Alabama at Birmingham (UAB) 2014. I have been on the UAB faculty since 2007. I received a BS in Chemistry at Pennsylvania State University in 1990 and a PhD in Chemistry from Michigan State University in 1998. I held a post-doctoral position at Argonne National Laboratory from 1999-2001, when I obtained the position of assistant professor at the University of Texas at El Paso.

In fact, the homepage at https://chemrxiv.org/engage/chemrxiv/public-dashboard clearly states at the top of the page “These are preliminary reports which have not been peer-reviewed. They should not be regarded as conclusive, guide clinical practice/health related behaviour, or be reported in news media as established information.”
Has this manuscript been submitted for publication and if so, where. What was the outcome of the peer review?
The authors make the claim that the enzymatic reactions that produce psilocin are “unexpected” for 5-MeO-DMT and therefore the synthesis of psilomethoxin is “unsubstantiated.” The authors are ignoring the 1988 study by Jochen Gartz where the transformation of diethyl‐tryptamine into 4‐hydroxy‐N,N‐diethyltryptamine (3.3%) and a minor quantity of 4‐phosphoryloxy‐N,N‐diethyltryptamine (≤0.8%) was produced by the Psilocybe cubensis mushroom in an example of biosynthesis of tryptamines. In addition, the authors do not cite Sherwood’s own research producing a very similar product via the reaction of 5‑Methylpsilocin to 5‑Methylpsilocybin, the very reaction they claim to be “unexpected.”[2]

The method used by Willaimson and Sherwood is not validated for the analysis of Psilomethoxin and 5-MeO-psilocybin and the authors have shown no evidence that their method would be able to identify Psilomethoxin and 5-MeO-psilocybin when it is present. The most important deficiency is that the method has never been tested with a certified sample of Psilomethoxin or 5-MeO-psilocybin.
A validated method would include the following:
Identification – known samples of Psilomethoxin or 5-MeO-psilocybin should be analyzed in order to determine that it is even possible to identify them with the proposed method.
A validated method will include the actual retention time and mass spectra for a known standard
Reproducibility – known samples of Psilomethoxin or 5-MeO-psilocybin should be analyzed multiple times on different days to show that the method can consistently identify these substances.
Limit of detection – known samples of Psilomethoxin or 5-MeO-psilocybin should be diluted and analyzed in order to determine the smallest concentration that can be identified by the method
Finally, while the authors claim they have no financial interest both are employed by USONA Institute https://www.usonainstitute.org which is currently conducting clinical trials in psilocin and 5-MeO-DMT and the authors areas of research include psilocin and 5-MeO-DMT. This indicates a strong financial interest in these potential therapeutics.
